The Plymouth Express follows the famous detective Hercule Poirot as he investigates a murder that takes place aboard the luxurious train named "The Plymouth Express". The episode begins with Poirot finding himself on the train heading for Plymouth when he stumbles upon a very delicate case. Whilst travelling on the train, he meets an American millionaire, Rufus Van Aldin, who takes a liking to Poirot and seeks his advice on a sensitive issue. Van Aldin is carrying a precious ruby that he wants to give to his daughter, Ruth Kettering as a wedding present. However, he is afraid that his daughter's husband, a notorious gambler named Derek Kettering, may try to steal it from her.
As the story progresses, tensions rise on the train as Van Aldin is found murdered, and the ruby is missing. As Poirot begins his investigation, he finds himself drawn into a complex web of lies and deceit. Ruth Kettering becomes a prime suspect, but Poirot is convinced that someone else is responsible.
Throughout the episode, there are a number of other characters who come under suspicion, including Kettering's mistress, his estranged brother-in-law, and the train's conductor. Poirot, with his brilliant deductive skills, questions each of the passengers and pieces together the clues to solve the case.
