In episode 3 of Miss Caretaker of Sunohara-sou, titled "Senpai + Change of Clothes + A Boy," the story follows Aki, a high school student who lives in Sunohara-sou, a dormitory for girls. Aki is struggling with his studies and decides to ask his senpai, Yuri, for help. Yuri is a smart and dependable girl, who is known for being strict with her juniors. When Aki visits her room to ask for help with his studies, she agrees to help him on one condition, that he wears female clothing as he studies.
At first, Aki refuses to go along with Yuri's condition, but eventually agrees to it when he realizes how serious she is about helping him. Yuri takes Aki to a nearby clothing store to buy some female clothing for him. Aki is reluctant at first, but Yuri manages to convince him that it's a necessary step for him to succeed in his studies. Once they return to Sunohara-sou, Yuri trains Aki on proper feminine mannerisms.
Aki is initially hesitant about wearing female clothing in front of his male classmates, but he eventually accepts that it is part of his new routine. He also starts to enjoy his new feminine persona, and begins to feel more comfortable around Yuri. Aki's newfound confidence and feminine appearance catch the attention of the other residents of Sunohara-sou, who start to see him in a different light.
Meanwhile, another resident of Sunohara-sou, Ayaka, is tasked with looking after a young boy who is staying at the dormitory. The boy, named Masaki, is the son of Ayaka's friend and is staying at Sunohara-sou for a few days. Masaki is young and energetic, and Ayaka struggles to keep up with him. She asks the other residents of Sunohara-sou for their help, and they all pitch in to look after Masaki.
As the day goes on, Aki is forced to switch back to his male clothing when he goes to his part-time job. However, he finds that he misses his female persona as he studies. This leads him to realize that he has developed feelings for Yuri and that he wants to be closer to her. Aki decides to continue wearing female clothing as he studies, hoping that it will bring him closer to Yuri.
In the end, Ayaka manages to look after Masaki for the day, and he leaves Sunohara-sou with fond memories of his stay. Aki continues to wear female clothing as he studies and makes progress in his studies. He also starts to open up to Yuri, who begins to see him in a new light.
