After a heated argument, Gintoki and Hijikata, the former Shinsengumi vice-commander, find themselves handcuffed to each other. The only way they can remove the cuffs is by finding the key, which somehow ended up in the hands of a woman named Ayame Sarutobi. Ayame is the leader of the Oniwabanshuu, a group of ninja who serve as the espionage and covert operations division of the Shogunate. She has a personal vendetta against the Shinsengumi, who she believes is responsible for her father's death.
Meanwhile, the rest of the gang tries to figure out a way to remove the cuffs without having to involve Ayame. Otae suggests they go to a blacksmith to cut them off, but Gintoki refuses, saying that it's impossible and the cuffs are made of some kind of unbreakable metal. Kagura decides they should try to make the best of the situation and come up with a plan to get Hijikata to cooperate, while Shinpachi frets over the fact that they're running late for work.
As Gintoki and Hijikata try to find Ayame, they encounter several obstacles in the form of Oniwabanshuu members. During one of the fights, Hijikata realizes that he and Gintoki are not so different after all. They both have a strong sense of duty and loyalty, and they both care deeply about their respective friends. However, their stubbornness and pride often get in the way of their ability to work together.
Meanwhile, Ayame confronts the Shinsengumi, and demands to know why they killed her father. The truth is revealed, and it turns out that her father sacrificed himself to save Hijikata's life. Ayame is shocked and saddened by this revelation, and she begins to question her motives.
Eventually, Gintoki and Hijikata find Ayame and engage in a fight with her. However, they are ultimately able to convince her to work together with them to take down a common enemy. With Ayame's help, they are able to retrieve the key and remove the cuffs.
In the end, Gintoki and Hijikata come to an understanding, and they both realize that their similarities and differences make them stronger when they work together. Ayame also learns to let go of her grudge against the Shinsengumi and begins to see them in a different light.
