In Scooby-Doo! in Where's My Mummy?, Scooby-Doo and the gang embark on another mystery-filled adventure. This time, the Mystery Inc. gang finds themselves in Egypt, where they are attempting to solve the case of the missing tomb of Cleopatra. The search for the tomb leads them to a modern-day amusement park, and the gang soon realizes that they are in for more than they bargained for.

The movie begins with a flashback to ancient Egypt, where we learn the legend of Queen Cleopatra's hidden tomb. The story goes that Cleopatra was buried with a great treasure, but her tomb was cursed to keep it hidden forever. Fast forward to modern times, and a group of archaeologists led by Professor Pericles (voiced by Ron Perlman) have found what they believe to be the location of Cleopatra's tomb. However, when they attempt to excavate the site, strange things begin to happen, and Pericles goes missing.

This is where the Mystery Inc. gang comes in. They receive a mysterious invitation to join an all-expenses-paid trip to Egypt, where they are promised a chance to solve the case of the missing tomb. Once they arrive in Egypt, they are greeted by a creepy figure known only as the High Priest, who warns them to stay away from the tomb.

Of course, being the brave and curious teenagers that they are, the gang quickly starts investigating. They soon discover that the entire trip is a ruse, and that they are actually in an amusement park modeled after ancient Egypt. The park is run by a man named Mr. Kubat (voiced by Ajay Naidu), who has a vested interest in the success of the archaeologists' excavation. Mr. Kubat and his henchmen are determined to keep the tomb hidden, and they will stop at nothing to make sure that the Mystery Inc. gang fails in their investigation.

As the gang continues to explore the amusement park and search for clues, they encounter a number of obstacles and enemies. They are chased by mummies, attacked by giant scorpions, and even trapped in a maze. Throughout the chaos, the gang tries to piece together the clues and unravel the mystery of Cleopatra's tomb.

Along the way, we get a glimpse into the personal lives of each member of the Mystery Inc. gang. Fred (voiced by Frank Welker) is as obsessed with traps and gadgets as ever, while Velma (voiced by Mindy Cohn) is her usual analytical self. Daphne (voiced by Grey Griffin) is determined to prove that she is more than just a pretty face, and Shaggy (voiced by Casey Kasem) and Scooby-Doo (also voiced by Welker) are up to their usual antics, scarfing down Scooby Snacks and stumbling into trouble.
